چکیده

A study of complementing lincomycin nonproducing mutants of Streptomyces lincolnensis resulted in the discovery of a low molecular weight compound that functions as a catalytic agent, lincomycin cosynthetic factor (LCF), in the biosynthesis of propylproline.^ This note reports the isolation and identification of LCF and its detection in fermentation broths of many actinomycetes. For the purification of LCF, 5. lincolnensis mutant NTG-5 fermentation broth was adjusted to pH 3.0, filtered, passed over an Diaion HP-20 chromatography column and eluted with a H2O MeOHgradient. Active fractions, identified by bioassay,1} were pooled, adjusted to pH 9.0, absorbed to Amberlite IRA 458 (OH") resin, and eluted with H2O1 m AcOH. The Diaion HP-20 column chromatography and later steps were also followed by TLC (silica plates developed with methyl ethyl ketoneacetone-water, 71 :19:10, Rf 0.41) with long

A robust synthesis of 7,8-didemethyl-8-hydroxy-5-deazariboflavin

The biosynthetic precursor of redox cofactor F420, 7,8-didemethyl-8-hydroxy-5-deazariboflavin, was prepared in four steps from 6-chlorouracil, 2-chloro-4-hydroxybenzaldehyde and bis-isopropylidene protected D-ribose. The latter aldehyde was transformed to the corresponding protected ribitylamine via the oxime, which was submitted to reduction with LiAlH4. Key advantage compared to previous synt...

Identification and cloning of the gene involved in the final step of chlortetracycline biosynthesis in Streptomyces aureofaciens.

For chlortetracycline biosynthesis in Streptomyces aureofaciens, the final reduction step is essential to give an antibiotic activity to its intermediate, which is catalyzed by tetracycline dehydrogenase with 7,8-dedimethyl-8-hydroxy-5-deazariboflavin (FO) as a cofactor. We identified and cloned the gene, which is essential for the biosynthesis of 6-demethyltetracycline and participates in the ...

An Efficient Intergeneric Conjugation of DNA from Escherichia coli to Mycelia of the Lincomycin-Producer Streptomyces lincolnensis

Streptomyces lincolnensis is a producer of lincomycin, which is a lincosamide antibiotic for the treatment of infective diseases caused by Gram-positive bacteria. S. lincolnensis is refractory to introducing plasmid DNA into cells because of resistance of foreign DNAs and poor sporulation. In this study, a simple and efficient method of transferring plasmids into S. lincolnensis through the int...

New Kid on the Block: LmbU Expands the Repertoire of Specialized Metabolic Regulators in Streptomyces.

Streptomyces has an extensive natural product repertoire, including most of the naturally derived antibiotics. Understanding the control of natural product biosynthesis is central to antibiotic discovery and production optimization. Here, Hou et al. (J. Bacteriol. 200:00447-17, 2018, https://doi.org/10.1128/JB.00447-17) report the identification and characterization of a novel regulator-LmbU-th...
